Buserelin Depot is an artificially created hormonal preparation of gonadotropin-releasing hormone. In most cases, it is used to suppress the synthesis of sex hormones , which in excess amounts can lead to endometriosis, fibroids or breast cancer.

Buserelin is used for the complex treatment of hormone-dependent malignant tumors in men and women. This product is available in the form of a nasal spray or solution for intramuscular injection. In the pharmacy, the medicine may be called: “Buserelin-depot”, “Buserelin-long” or simply “Buserelin”. This synthetic analogue of natural gonadotropin-releasing hormone is especially useful in the treatment of endometriosis (the main complication of which is infertility).

Buserelin is a hormonal drug with antiandrogenic, antigonadotropic and antiestrogenic properties. As already mentioned, it has an antitumor effect by binding to the receptors of pituitary cells, causing a short-term increase in the level of hormones in the blood. By reducing the synthesis of testosterone, it has a suppressive effect on the synthesis of sex hormones . With a continuous course, the drug reduces inflammatory processes, the size of pathological tissue, and reduces its blood supply.

Some patients do find it effective, but many claim that certain unwanted reactions, including cysts, have occurred after use. "Buserelin", according to doctors, can only be used under the qualified supervision of the attending physician. However, it is often used in medical practice and helps in the treatment of uterine fibroids, hyperplastic endometrial processes and endometriosis, as well as tumor hormone-dependent processes of the genital area in women.

How to take Buserelin for endometriosis?

First of all, it is worth noting that the drug is a suspension for intramuscular administration . Buserelin is diluted right before injection . The ampoule comes with two needles with different colored caps. It is very important not to confuse their sequence , because the quality of the injection depends on this. A needle with a pink tip is used to draw up a solvent and inject it into an ampoule with a dry lyophilisate. The suspension is shaken and drawn into a syringe in a vertical position. After this, the needle is replaced with another one with a green tip and the injection is performed.

For endometriosis, Buserelin is administered once, at a dosage of 4.2 mg four times a month. As a rule, injections begin in the first five days of the menstrual cycle.

The course of treatment can range from 3-6 months . After this time, the patient is prescribed a general blood test and a hormone test, and a pelvic ultrasound is performed.

After the first injection, I didn’t notice any side effects, but my periods came, heavy, as usual. But after the second, strong hot flashes began. At night I didn’t sleep, I had a terrible headache, I tried all the pills for headaches, only Pentalgin helped. I lost 2 kg in a week, because... I felt sick and couldn’t eat. My blood pressure began to rise significantly, it reached 194/121, my pulse was under 100. I started taking Prestarium 5 mg. At night I had dry mouth, which caused a cough, my periods came scanty. After the third injection, the side effects were the same , nagging pain in the lower abdomen increased and my lower back began to hurt. I noticed a change in mood. Sometimes I wanted to cry for no clear reason. I went to the doctor, an ultrasound showed that the endometrium was normal, the fibroids remained the same. They prescribed Femoston 5 mg + 1 mg. once a day. After the third injection, my periods came VERY heavy, lasted for a week. I took the fourth injection, while taking Femoston, the side effects no longer tormented me, but the joints on my hands began to hurt. My periods came again, but they were more scanty, the duration was the same I had it for 7 days, there was a turmoil in my stomach, I’m still trying to cope with this with Linex, 6 tablets a day. Now I took the 5th injection, more pain in the joints, especially in the shoulders, drowsiness, it’s very difficult to get up in the morning, but no one canceled work. I’m waiting, what will happen next...they will come, they won’t come.....I don’t even know what to say yet, whether it’s worth using it or not. But it’s for sure that doctors need to take it more seriously and be more attentive to the patients who are prescribed this drug. that before this you need to undergo some additional tests, I’m not a doctor, I don’t know. I read a lot about this drug, about the reviews, I see that very often problems begin in the mammary glands, often leading to operations. I also have pain, but I’ve had fibrocystic mastopathy for a long time, I’m being monitored, but after the sixth injection I want to get checked unscheduled, on my own initiative. The principle of action of the drug

The principle of action of the drug is due to its ability, as a gonadotropin-releasing hormone agonist, to suppress the function of gonadotropic hormones, in particular follicle-stimulating hormone and luteinizing hormone, which regulate the production of estrogen. Buserelin depot competitively binds to the receptors of the pituitary lobe, activating a short-term increase in the level of sex hormones, which is observed during the first 10 days of administration.

After such an outbreak, the receptors of the cells of the anterior pituitary gland, which produce gonadotropic hormones, completely lose sensitivity to the stimulating effects of the drug. As a result, the secretion of gonadotropic hormones decreases, and with continued use of Buserelin Depot, it is completely blocked.

As a result, on the 12th–14th day of taking the drug, there is a complete suppression of the synthesis of sex hormones in the ovary and a decrease in the concentration of estradiol in the blood to postmenopausal values.
